Ok, I've just gone through and checked everything. I checked the templates, removed and reapplied the languages, tested it with and without browser language and URL language detection and also restarted the app and validated the store as Dave mentioned.
At this point it's working with US and IT enabled. You might need to get on there quick and disable US again if it's not meant to be enabled.
One suggestion when you're setting things up is to always run it with debug mode enabled in the back office. That saves you having to restart the app when making certain changes.
