I may have a couple of tips to try...
When was the last time you rebooted the server?
Have you tried to run the app pool with 32 bit enabled?
What worker process are you using, network service?
If you use dnn7 you need to run the app pool with asp 4.0, required by dnn.
Is asp.net installed correctly?
